We have developed a set of novel efficient transition metal-catalyzed methodologies for synthesis of heterocycles. Generally, regioselective synthesis of multisubstituted heterocycles is a challenging task. However, we have shown that incorporation of migrating step(s) in the cyclization sequence often helps solving this problem. Thus, it was found that, in the presence of Cu-, Ag-, and Au catalysts, a number of groups, such as Hal-, RS-, AcO-, TsO-, and Ar- could undergo 1,2- or 1,3-migration, which allows for expeditious synthesis of densely-substituted heterocycles, which are not easily accessible via existing methodologies.

We have also developed novel Cu-catalyzed tri-component coupling methods for highly convergent and efficient synthesis of indolines, indoles, and imidazopyridines, valuable heterocyclic cores, widely found in pharmacologically important molecules.

We have also explored a direct Pd-catalyzed C-H functionalization approach toward synthesis of multisubstituted aromatic and heteroaromatic molecules. Moreover, employment of a silicon-tether motif opens new opportunities for a direct or directed functionalization of aromatic molecules via a Pd-catalyzed C-H activation pathway. The scope of these transformations will be demonstrated and the mechanisms will be discussed.
